An Ayurvedic prospective of Manjishtha

Though Rubia cordifolia has property of bitter, astringent and sweet in taste and heavy, dry and hot in effect, it is a great agent to lower down kapha and pitta. Besides the glucosides known as manjisthin and purpurine, it has chemical composition that consists of many other components, which include resins, lime salts and coloring agents.

The valuable herb Manjishtha has proven efficacy of treating a number of diseases. It is a drug of choice for treating various systemic problems like elevated uric acid and gouty arthritis, glandular swellings, recurrent skin infections. Especially for diseases of the skin like pigmentation anomalies and leucoderma it has significant positive effects.

Rubia cordifolia has been used in various formulations to treat uterine and urinary infections. The herb also shows good effect on soothing gastrointestinal track and hence can be used for some of the digestive problems such as diarrhea and dysentery. The herb is also used to treat chronic fevers in ancient text of Ayurveda.

Manjishtha holds the quality of a very good herb for the skincare. It can be used externally and internally. A capsule made out of its extract can help one to gain luster and glow (of the skin) and aids to remove pimples, freckles and discoloration or pigmentation.


Action wise researches of Madder or Rubia cordifolia

The roots are alterative, anodyne, antiphlogistic, antitussive, astringent, and diuretic, emmenagogue, expectorant, styptic, tonic and vulnerary [1, 2, 3, 4, and 5].

They have an antibacterial action, inhibiting the growth of Staphylococcus aureus, S. epidermidis, Pneumococci etc. They are used to lower the blood pressure [2].

The roots have been used internally in the treatment of abnormal uterine bleeding, internal and external hemorrhage. They are also used in the treatment for bronchitis, rheumatism, stones in the kidney, bladder and gall, dysentery etc. The roots are harvested in the autumn from plants that are at least 3 years old. They are peeled and then dried and then could be used in the form of extract [6].

The stem of the herb Manjishtha is used in Tibetan medicine, where they are said to have a bitter taste and a cooling potency Febrifuge, they are used in the treatment of blood disorders and spreading fever of kidneys and intestines [7].

Other clinical studies of Madder

Effect of alcoholic extract of roots of Rubia cordifolia was trialed on elevated blood glucose level in alloxan treated animals. The extract reduced the blood glucose level raised by alloxan. Effect of alcoholic extract was also investigated on cold restraint induced stress and on scopolamine-induced memory impairment. Alcoholic extract enhanced brain gamma-amino-n-butyric acid (GABA) levels and decreased brain dopamine and plasma corticosterone levels. Acidity and ulcers caused due to cold restraint stress were inhibited by alcoholic extract. Animals treated with alcoholic extract spent more time in open arm in elevated plus maze model. It also antagonized scopolamine induced learning and memory impairment. In some study, ethanol, methanol, ethyl acetate and water extracts of Rubia tinctorum L. (Rubiaceae) were tested for antimicrobial activity by the disc diffusion method. From the present study it was found that Rubia tinctorum L. revealed antimicrobial activity against some Gram (+) and Gram (-) bacteria, yeasts, filamentous fungi and actinomycetes - ( Phytother Res. 2006 Jun;20(6):490-2. Research on wound healing drugs is a developing area in modern biomedical sciences. Scientists who are trying to develop newer drugs from natural resources are looking toward the Ayurveda, the Indian traditional system of medicine. Several drugs of plant, mineral, and animal origin are described in the Ayurveda for their wound healing properties under the term Vranaropaka. Most of these drugs are derived from plant origin. Some of these plants have been screened scientifically for the evaluation of their wound healing activity in different pharmacological models and patients, but the potential of most remains unexplored. In a few cases, active chemical constituents were identified. Some Ayurvedic medicinal plants, namely, Ficus bengalensis, Cynodon dactylon, Symplocos racemosa, Rubia cordifolia, Pterocarpus santalinus, Ficus racemosa, Glycyrrhiza glabra, Berberis aristata, Curcuma longa, Centella asiatica, Euphorbia nerifolia, and Aloe vera, were found to be effective in experimental models. The major damaging factor during and after the ischemic/hypoxic insult is the generation of free radicals, which leads to apoptosis, necrosis and ultimately cell death. Rubia cordifolia (RC), Fagonia cretica linn (FC) and Tinospora cordifolia (TC) have been reported to contain a wide variety of antioxidants and have been in use in the eastern system of medicine for various disorders. However, their mechanism of action was largely unknown. We therefore selected these herbs for the present study to test their neuroprotective ability and the associated mechanism in rat hippocampal slices subjected to oxygen-glucose deprivation (OGD). The results revealed that all the three herbs were effective in elevating the GSH levels, expression of the gamma-glutamylcysteine ligase and Cu-Zn SOD genes. The herbs also exhibited strong free radical scavenging properties against reactive oxygen and nitrogen species as studied by electron paramagnetic resonance spectroscopy. In addition all the three herbs significantly diminished the expression of iNOS gene after 48 hours which plays a major role in neuronal injury during hypoxia/ischemia.


The Manjishtha plant

The Common Madder can grow to 1.5 m in height. The evergreen leaves are 5-10 cm long and 2-3 cm broad, produced in whorls of 4-7 star like around the central stem. It climbs with tiny hooks at the leaves and stems. The flowers are small (3-5 mm across), with five pale yellow petals, in dense racemes, and appear from June to August, followed by small (4-6 mm diameter) red to black berries. The roots can be over a meter long, up to 12 mm thick and the source of a red dye known as rose madder. It prefers loamy soils with a constant level of moisture.


Manjishtha ’s effect on free radicals:

The major negative factor during and after the ischemic/hypoxic insult is the generation of free radicals those lead to apoptosis, necrosis, and ultimately cell death. Rubia cordifolia (RC) has been reported to contain a wide range of antioxidants and have been in practice in the ancient eastern system of medicine to cure various disorders. Hippocampal slices were subjected to oxygen-glucose deprivation (OGD) and divided into 3 groups namely control, OGD, and OGD+drug treated. Cytosolic reduced glutathione (GSH), nitric oxide [NO, measured as nitrite (NO 2)]. EPR was used to establish the antioxidant action of RC with respect to superoxide anion (O 2 .-), hydroxyl radicals ( .OH), nitrite oxide (NO) radical, and peroxynitrite (ONOO -) generated from pyrogallol, menadione, DETA-NO, and Sin-1, respectively. RT-PCR was performed for the herb to assess its effect on the expression of γ-glutamylcysteine ligase (GCLC), iNOS, and GAPDH gene expression.

At the end of the experiment, it was found that Manjishtha was effective in elevating the GSH levels and expression of the GCLC. The herb also exhibited strong free radical scavenging properties against reactive oxygen and nitrogen species as carried out by electron paramagnetic resonance spectroscopy, diminishing the expression of iNOS gene. RC, therefore attenuate oxidative stress mediated cell injury during OGD and exert the above effects at both the cytosolic as well as at gene expression degrees and may be effective therapeutic tool against ischemic brain damage.
